چکیده

Aim and methods We generated DCleu ex vivo from AML blasts from heparinised whole blood (‘WB-DC’, to simulate the in vivo situation) from 65 AML-pts in active stages of the disease using standard methods ( ‘Picibanil’, ‘MCMMimic’, ‘Ca-ionophore’, ‘IFNa’) or 11 minimalised cocktails (‘WB-minicock-DC’, combinations of 1-3 selected cytokines, antibiotics, bacterial lysates, or other clinically approved response-modifiers) and to correlate proportions of DCor T cellsubsets and cytokine profiles with results with their ex vivo stimulatory capacity for antileukaemic T-cells and the pts’ response to immunotherapy (SCT/DLI).

Immunotherapy using bispecific T cell engager (BiTE®) antibodies: preclinical and clinical experience in acute leukemia

BiTE antibodies are novel recombinant single chain Ig domain constructs that leverage the endogenous cytotoxic potential of polyclonal T cells to target malignant cells by utilizing the specific binding properties of variable domains from two different antibodies. Antibody-based immunotherapy represents a promising strategy in cancer. BiTE antibodies have demonstrated efficacy in hematologic ma...

Up-regulation of anti-apoptotic genes confers resistance to the novel anti-leukaemic compound PEP005 in primary AML cells

We showed previously that PEP005 induced apoptosis in leukaemic cell lines and blasts from patients with acute myeloid leukaemia (AML). Here we assess the anti-leukeamic effects of PEP005 in vivo and determine the mechanism of resistance of PEP005 non-responsive cells. We used 2 human xenograft mouse models of AML to assess the anti-leukaemic effects of PEP005 in vivo. Expression microarray ana...

Preparation and characterization of a novel nanobody against T-cell immunoglobulin and mucin-3 (TIM-3)

Objective(s): As T-cell immunoglobulin and mucin domain 3 (TIM-3) is an immune regulatory molecule; its blocking or stimulating could alter the pattern of immune response towards a desired condition. Based on the unique features of nanobodies, we aimed to construct an anti-TIM-3 nanobody as an appropriate tool for manipulating immune responses for future therapeutic purposes. Materials and Meth...

Acute myeloid leukemia with T-lymphoid features: a distinct biologic and clinical entity.

We studied the clinical and biologic features of 10 cases of acute leukemia that met standard French-American-British (FAB) criteria for acute myeloid leukemia (AML) but in which the blast cells also expressed the T-cell-associated CD2 surface antigen. All cases had greater than 3% myeloperoxidase and Sudan black B-positive leukemic blasts, and blasts from seven cases contained Auer rods. React...

Clinical and Laboratory Findings of Cup-Like Nuclei in Blasts with FLT3 Mutation in Pediatric Acute Myeloid Leukemia: A Case Report

Biologically, Acute myeloid leukemia (AML) is highly heterogenous. AML with cup-like blast morphology variant has been reported to have important role in risk group stratification and treatment implications. In pediatric age group, this morphology and its clinical implication is rarely discussed. Although this morphology variant is not stated in World Health Organization (WHO) classification of...
